Summary and explanation of the test
Cancer Antigen 125 (CA-125) is a glycoprotein that occurs inblood as high molecular weight (Mr>200,000). High concentrationsof this antigen are associated with ovarian cancer and a range ofbenign and malignant diseases. Although the specificity andsensitivity of CA-125 assays are somewhat limited, especially inearly diagnosis of ovarian cancer, the assay has found widespreaduse in the differential diagnosis of adnexal masses, in monitoringdisease progression and response to therapy in ovarian cancer, andin the early detection of recurrence after surgery of chemotherapyfor ovarian cancer. Published literature has shown that elevatedserum CA-125 levels can be observed in patients with seriousendometriosis, clear cell and undifferentiated ovarian carcinoma.The serum CA-125 is elevated in 1% of normal healthy women, 3% ofnormal healthy women with benign ovarian diseases, and 6% ofpatients with non-neoplastic conditions.
The employment of several serum references of known CancerAntigen 125(CA-125) levels permits the construction of a doseresponse curve of activity and concentration. From comparison tothe dose response curve, an unknown specimen’s activity can becorrelated with CA-125 concentration.

Specimen collection and preparation
The specimens shall be blood serum in type and the usualprecautions in the collection of venipuncture samples should beobserved. For accurate comparison to established normal values, afasting morning serum sample should be obtained. The blood shouldbe collected in a plain red-top venipuncture tube with or withoutgel barrier. Allow the blood to clot. Centrifuge the specimen toseparate the serum from the cells. Samples may be refrigerated at2~8Degree Celsius for a maximum period of five days. If thespecimen can not be assayed within this time, the sample may bestored at temperatures of -20Degree Celsius for up to 30 days.Avoid repetitive freezing and thawing.
